Second harmonic imaging of membrane potential dyes. Signal arises only from asymmetrically labeled membrane regions (majority of the dye in the outer leaflet), increasing the signal-to-background ratio. Some reports of up to 60% change in SHG per 100 mV. However, in this case the SH is detected only in transmission mode, precluding use on thick samples (e.g. deep in a brain slice). Di-8-ANEPPS loaded RBL cells: Red pseudo-color is fluorescence; green is SHG. Exc. was at 920 nm.
